3d Hemogenic Gastruloid

A 3D hemogenic gastruloid is a stem cell-derived, three-dimensional model that recreates key features of early embryonic development and the emergence of blood-forming cells, providing a platform for studying cancer-related hematopoiesis. Aggregated pluripotent stem cells self-organize into patterned tissues, where signaling cues establish germ layers and induce hemogenic endothelium, the specialized vascular tissue that generates hematopoietic progenitors through endothelial-to-hematopoietic transition. In cancer research, these gastruloids can help investigate how developmental pathways become disrupted in leukemia and other blood disorders, model oncogenic changes in human cell populations, and evaluate potential therapies in a physiologically relevant developmental context. Their controlled, scalable organization also supports comparative studies of normal and malignant blood-cell formation.

Hematopoietic stem and progenitor cells (HSPC) derive from specialized (hemogenic) endothelial cells during development, yet little is known about the process by which some endothelial cells specify to become blood forming. We demonstrate a flow-cytometry based method allowing simultaneous isolation of hemogenic endothelial cells and HSPC from murine embryonic tissues.

The protocol described here details the induction of a hemogenic program in mouse embryonic fibroblasts via overexpression of a minimal set of transcription factors. This technology may be translated to the human system to provide platforms for future study of hematopoiesis, hematologic disease, and hematopoietic stem cell transplant.

Invasion Assay Using 3D Matrices

The extracellular matrix (ECM) is a network of molecules that provide a structural framework for cells and tissues and helps facilitate intercellular communication. Three-dimensional cell culture techniques have been developed to more accurately model this extracellular environment for in vitro study. While many cell processes during migration through 3D matrices are similar to those required for movement across rigid 2D surfaces, including adherence, migration through ECM also requires cells...
