Ev Elution

EV elution is the process of recovering extracellular vesicles (EVs) from a separation or affinity-capture material while preserving their structure and molecular cargo. Elution typically releases bound vesicles by changing conditions such as pH, ionic strength, or ligand competition, allowing EVs to detach from the capture surface and enter a collection solution. In cancer research, efficient elution supports the isolation of tumor-associated EVs for analysis of proteins, lipids, and nucleic acids, including circulating biomarkers. Recovered vesicles can also be used in studies of intercellular communication, disease progression, treatment response, and potential diagnostic or therapeutic applications.

Here, we present a protocol to evaluate the antibacterial efficacy of an antibiotic-eluting polymer to simulate prophylactic clinical application by using a commercially available real-time ATP-based luminescent microbial viability assay. This method enables the monitoring of the longitudinal activity of drug-eluting materials and can be widely adapted to test anti-microbial drug delivery platforms.
