Quantitative Dna Assessment

Quantitative DNA assessment is the measurement of DNA amount or content in a biological sample, providing a numerical basis for evaluating genome status and sample quality. Depending on the assay, DNA is extracted and measured through absorbance or fluorescence, while cellular DNA content can be compared with reference standards to identify changes in ploidy and the distribution of cells across the cell cycle. In cancer research, these measurements support tumor characterization, detection of abnormal genome content, assessment of genomic instability, and evaluation of treatment-associated changes. Reliable quantification also helps normalize downstream molecular analyses and improve comparisons across samples and experiments.

Here, we present a protocol for tracing genomic DNA (gDNA) contamination in RNA samples. The presented method utilizes primers specific for the internal transcribed spacer region (ITS) of ribosomal DNA (rDNA) genes. The method is suited for reliable and sensitive detection of DNA contamination in most eukaryotes and prokaryotes.

We provide a simple, semi-quantitative method to investigate biofilm formation in vitro. This method takes advantage of the Zeiss stemi 2000-C Dissecting Microscope (with camera attachment) to monitor both the timing and pattern of biofilm formation, as assessed by the development of wrinkled colonies.
