Methyl Acetate

Methyl acetate is a volatile organic compound and ester with the formula CH₃COOCH₃, formed from acetic acid and methanol and valued as a solvent in chemistry and manufacturing. Its ester functional group contains a carbonyl carbon bonded to an oxygen-linked methyl group, allowing the compound to participate in nucleophilic acyl substitution, including acid- or base-catalyzed hydrolysis that produces acetic acid or acetate and methanol. Methyl acetate dissolves many organic substances while evaporating readily, supporting its use in coatings, adhesives, inks, and extraction processes. Studying its structure and reactivity illustrates key principles of ester chemistry, solvent selection, and chemical equilibrium.

A method for the determination of acetate kinase activity is described. This assay utilizes a direct reaction for determining enzyme activity and kinetics of acetate kinase in the acetate-forming direction with different phosphoryl acceptors. Furthermore, this method can be utilized for assaying other acetyl phosphate or acetyl-CoA utilizing enzymes.

DNA Methylation Analysis

2023

Methylation at CpG dinucleotides is a chemical modification of DNA hypothesized to play important roles in regulating gene expression. In particular, the methylation of clusters of methylation sites, called “CpG islands”, near promoters and other gene regulatory elements may contribute to the stable silencing of genes, for example, during epigenetic processes such as genomic imprinting and X-chromosome inactivation. At the same time, aberrant CpG methylation has been shown to be associated with...

DNA Methylation: Bisulphite Modification and Analysis

The gold standard for DNA methylation analysis is genomic sequencing of bisulphite converted DNA. This method takes advantage of the increased sensitivity of cytosine compared with 5-methylcytosine (5-MeC) to bisulphite deamination under acidic conditions. Unmethylated cytosines can be distinguished from methylated cytosines after PCR amplification of the target genomic DNA.

The ruthenium-catalyzed olefination of electron-deficient alkenes with allyl acetate is described here. By using aminocarbonyl as a directing group, this external oxidant-free protocol has high efficiency and good stereo- and regioselectivity, opening a novel synthetic route to (Z,E)-butadiene skeletons.
