Protein Peptide Analysis

Protein peptide analysis is the chemical characterization of proteins and their constituent peptides to determine composition, structure, sequence, and modifications. It commonly combines enzymatic digestion or chemical hydrolysis with separation methods such as liquid chromatography and detection by mass spectrometry, where peptide mass-to-charge ratios and fragmentation patterns support sequence identification. In chemistry, these analyses help verify synthetic and purified biomolecules, map post-translational modifications, assess protein purity, and investigate interactions or structural changes. The resulting molecular information supports pharmaceutical quality control, biochemical research, biomarker studies, and the development of protein-based therapeutics.

Biotinylated Cell-Penetrating Peptide Probe to Detect Protein-Protein Interactions

2025

This video demonstrates a technique that uses biotinylated cell-penetrating peptides to detect intracellular protein-protein interactions. The cell-penetrating peptides facilitate the conjugated peptide to enter a cell and specifically bind to its interacting proteins. The biotin, after binding to avidin, allows purification of the interacting proteins’ complex from a cell lysate.

In this video, we demonstrate reversed-phase high-performance liquid chromatography to purify peptides from a digested protein lysate mixture. These purified peptides are then lyophilized and stored for downstream applications. Analysis of peptides can help understand the mechanics involved in cellular signaling and cancer.
