Immunological Cell Sorting

Immunological cell sorting is the selective isolation of immune-cell populations from a mixed sample according to surface markers, physical properties, or functional state, enabling precise analysis of heterogeneous tissues. In fluorescence-activated cell sorting (FACS), fluorescently labeled antibodies bind defined antigens, and a flow cytometer measures fluorescence and light scatter before sorting labeled cells into separate droplets; magnetic-activated cell sorting (MACS) uses antibody-coated magnetic beads and a magnetic field for enrichment or depletion. These approaches support the study of lymphocyte subsets, infection-driven immune responses, cell-cell interactions, and rare populations. Purified cells can then undergo culture, molecular profiling, or functional assays.

Immunological Synapse Formation Between Helper T Cells and Antigen-Presenting Cells

This video demonstrates the in vitro study of immunological synapse formation between an antigen-presenting cell and helper T lymphocytes using a fluorescence microscope. T cell receptors of helper T lymphocytes interact with an antigen-MHC-II complex in antigen-presenting cells and form immunological synapses, followed by secretory vesicle movements toward the synapse, which is visualized by fluorescent microscopy.
