Orthotopic Graft Placement

Orthotopic graft placement is a transplantation technique in which donor tissue, cells, or an organ is positioned at the recipient’s corresponding anatomical site, preserving relevant local structure and function. The graft interacts with the surrounding microenvironment through tissue contact, vascular integration, and immune recognition, while compatibility, inflammation, and infection control influence its establishment. In immunology and infection research, orthotopic models help investigators examine rejection, pathogen behavior, host defenses, and immune responses under physiologically relevant conditions. These models can improve understanding of graft survival and disease progression and support evaluation of immunosuppressive strategies, vaccines, and antimicrobial interventions.

Two techniques can be used to establish this model: injection of a cancer cell suspension into the cecal wall or transplantation of a piece of subcutaneous tumor onto the cecum.
