Prp C Denaturation

PrP C denaturation is the loss of the native three-dimensional structure of the cellular prion protein, PrPᶜ, a process important for understanding prion-related disease. Changes in temperature, pH, or exposure to chaotropic agents disrupt the noncovalent interactions that stabilize PrPᶜ, exposing hydrophobic regions and potentially promoting conversion into misfolded, beta-sheet-rich aggregates associated with PrPˢᶜ. Studying this transition helps researchers characterize prion protein stability, aggregation, and infectivity at the molecular level. Denaturation models also support the development of biochemical assays, clarify mechanisms of neurodegeneration, and guide investigations of compounds that could prevent or reverse pathogenic protein misfolding.

Implant-associated infection is a significant clinical complication. This study describes an approach using platelet-rich plasma (PRP) to prevent implant-associated infections, presents the protocol for preparing PRP with constant platelet concentration, and reports the newly identified antimicrobial properties of PRP and related protocols for examining such antimicrobial properties in vitro.

A new species of cellular prion protein (PrPC) has recently been identified in uninfected human brains using the methods described here. These methods can be used to isolate various PrP species, while some of them are also useful in isolating other misfolded protein aggregates from human brains.
