Acute Slices

Acute slices are freshly prepared sections of nervous tissue maintained outside the organism for a limited period, providing a practical model for studying neuronal circuits under controlled conditions. Tissue is rapidly dissected, cut in oxygenated physiological solution, and kept viable while neurons retain much of their local synaptic connectivity and membrane function. Researchers combine acute slices with patch-clamp recording, field potentials, stimulation, imaging, or pharmacology to examine synaptic transmission, excitability, plasticity, and circuit interactions. Because the preparation offers experimental access while preserving native microcircuit features, it supports investigations of normal brain function, disease mechanisms, and the effects of candidate treatments.

Preparation of Acute Subventricular Zone Slices for Calcium Imaging

A method to load subventricular zone (SVZ) cells with calcium indicator dyes for recording calcium activity is described. The postnatal SVZ contains tightly packed cells including neural progenitor cells and neuroblasts. Rather than using bath loading we injected the dye by pressure inside the tissue allowing better dye diffusion.

Imaging Pheromone Sensing in a Mouse Vomeronasal Acute Tissue Slice Preparation

In mice, the ability to detect pheromones is principally mediated by the vomeronasal organ (VNO). Here, an acute tissue slice preparation of VNO for performing calcium imaging is described. This physiological approach allows observations of subpopulations and/or individual neurons in a living tissue and is convenient for receptor-ligand identification.

Time-lapse Imaging of Neuroblast Migration in Acute Slices of the Adult Mouse Forebrain

We describe a protocol for real-time videoimaging of neuronal migration in the mouse forebrain. The migration of virally-labeled or grafted neuronal precursors was recorded in acute live slices using wide-field fluorescent imaging with a relatively rapid acquisition interval to study the different phases of cell migration, including the durations of the stationary and migration phases and the speed of migration.

In this paper we show a method for preparing acute brain slices in physiological temperature, using a conventional physiological solution without special modifications for the cutting (such as adding sucrose) and without intracardial perfusion of the animal before slice preparation.
