Awake Rat Injection

Awake rat injection is a neuroscience technique for administering drugs, tracers, or other experimental substances to a conscious rat, allowing researchers to study neural function without anesthesia-related effects. The procedure uses controlled restraint and a carefully positioned needle or catheter to deliver material through a selected route, such as systemic or localized injection, while maintaining animal welfare and minimizing movement. Researchers use this approach to examine behavioral responses, pharmacological effects, neural signaling, and circuit function in real time. By preserving wakeful brain activity, awake rat injection can improve the physiological relevance of experiments involving cognition, motivation, pain, and other behaviorally expressed processes.

Electrophysiological characterization of neuronal responses is important for understanding brain function and for guiding the placement of dyes for pathway tracing. However, many studies are performed in anesthetized animals. To understand brain function without anesthetics, we developed a method to record neuronal response properties and inject dyes in awake mouse.

(2015)This video demonstrates the method of juxtacellular recording and staining in a restrained, awake rat. The procedure involves recording signals from neurons and injecting dye to precisely localize the recording site. This technique allows accurate mapping of neural activity to the brain's anatomical structure.
