Combined Infusion Stimulation

Combined infusion stimulation is a neuroscience approach that pairs the controlled delivery of a drug or other solution with electrical or sensory stimulation to examine how chemical and neural inputs interact. The infused compound alters signaling at targeted receptors, circuits, or synapses, while stimulation produces a measurable neural or behavioral response under defined experimental conditions. Comparing responses with and without the infusion can reveal drug effects, receptor involvement, and circuit mechanisms. This combined design supports research on neurotransmission, neuromodulation, learning, and neurological disorders by linking pharmacological changes to functional activity in nervous system pathways.

We present a microinjectrode system designed for electrophysiology and assisted delivery of experimental probes (i.e., nanosensors, microelectrodes), with optional drug infusion. Widely available microfluidic components are coupled to a cannula containing the probe. A step-by-step protocol for microinjectrode construction is included, with results during muscimol infusion in macaque cortex.

Transcranial Alternating Current Stimulation (tACS) allows the modulation of cortical excitability in a frequency-specific fashion. Here we show a unique approach which combines online tACS with single pulse Transcranial Magnetic Stimulation (TMS) in order to "probe" cortical excitability by means of Motor Evoked Potentials.
