Cylinder Testing Method

The cylinder testing method is a behavioral assay that measures spontaneous forelimb use and motor asymmetry in rodents, making it valuable for assessing neural injury and recovery. During the test, an animal explores a transparent vertical cylinder, and researchers score the forepaws used for wall contact, support, and movement; unequal use can indicate impaired motor control on one side of the body. In neuroscience, the method helps evaluate functional deficits after stroke, traumatic brain injury, or other experimental interventions, while repeated testing can track progression, treatment effects, and behavioral recovery alongside anatomical or molecular findings.

Classical forelimb asymmetry analysis of the cylinder test is routinely used to assess behavioural deficits in rats following brain injury or stroke; however, it fails to detect consistent deficits in mice. This study demonstrates that quantifying paw-dragging behaviour is a more sensitive analysis of brain injury in mice.

In this study, Er,Cr:YSGG and diode lasers applied separately to the flat surface of a total of 96 specially designed titanium cylinders. A thermocouple placed on the other surface and the temperature was measured. Surface roughness analyzed by profilometer, SEM and AFM.
