Permeability Measurements

Permeability measurements quantify how readily a substance passes through a material, membrane, or biological barrier, making them essential for evaluating transport in bioengineering systems. In a typical assay, a concentration or pressure gradient drives molecules or fluid across a test barrier, while the resulting flux is measured to calculate a permeability coefficient under defined conditions. These measurements can assess solute transport through engineered membranes, biomaterials, cell layers, and tissue models. They support the design of drug-delivery devices, filtration systems, and tissue-engineered constructs, while helping researchers compare barrier integrity, material performance, and changes caused by structure or experimental conditions.

Multi-parameter Measurement of the Permeability Transition Pore Opening in Isolated Mouse Heart Mitochondria

A spectrofluorometric protocol for the measurement of the mitochondrial permeability transition pore opening in isolated mouse heart mitochondria is presented here. The assay involves the simultaneous measurement of mitochondria Ca2+ handling, mitochondrial membrane potential and mitochondrial volume. The procedure for obtaining high-quality and functional heart mitochondria is also described.

An Assay for Permeability of the Zebrafish Embryonic Neuroepithelium

We describe a live whole animal quantitative measurement for permeability of the embryonic zebrafish brain. The technique analyzes the ability to retain cerebrospinal fluid and molecules of different molecular weights within the neural tube lumen and quantifies their movement out of the ventricles. This method is useful for determining differences in epithelial permeability and maturation during development and disease.

Measuring the osmotic water permeability coefficient (Pf) of cells can help understand the regulatory mechanisms of aquaporins (AQPs). Pf determination in spherical plant cell protoplasts presented here involves protoplasts isolation and numerical analysis of their initial rate of volume change as a result of an osmotic challenge during constant bath perfusion.
