Arginine Deiminase Treatment

Arginine deiminase treatment is a cancer therapy that lowers circulating arginine, an amino acid required for protein synthesis and cell growth. The enzyme converts arginine into citrulline and ammonia, creating nutrient stress that can preferentially affect tumors with limited expression of argininosuccinate synthetase 1 (ASS1), which cannot efficiently resynthesize arginine. In cancer research, this approach is used to study tumor metabolic dependencies and evaluate selective strategies against arginine-auxotrophic cancers. Treatment outcomes depend on tumor biology, enzyme activity, immune effects, and resistance mechanisms, supporting the use of ASS1 status and related metabolic markers to guide combination therapies and clinical investigation.

Our studies have revealed that the beneficial effects of conivaptan are dependent on the method of delivery after experimental stroke in mice. We have developed a research protocol for delivery of the receptor blocker via IV catheter on stroke-evoked brain edema formation in mice.
