Cytokine-containing Patch

A cytokine-containing patch is a biomaterial-based platform designed to deliver cytokines locally and support controlled communication between cells. By retaining cytokines within or on a patch and releasing them at the target site, it can regulate signaling pathways that influence cell survival, proliferation, migration, and differentiation. In developmental biology, these patches provide a localized way to study how spatial and temporal patterns of cytokine signaling shape tissue formation and repair. They may also support engineered tissue models, organoid research, and regenerative strategies by creating defined signaling environments while limiting exposure to surrounding tissues.

Described here is a method for producing exogenous cytokine in patient-derived xenograft (PDX) mice via weekly intraperitoneal injection of a cytokine-transduced stromal cell line. This method broadens the utility of PDX and provides the option for transient or sustained exogenous cytokine delivery in a multitude of PDX models.

Neuron cell membranes are populated with ion channels that control the movement of charge into and out of the cell, thereby regulating neuron firing. One extremely useful technique for investigating the biophysical properties of these channels is called patch clamp recording. In this method, neuroscientists place a polished glass micropipette against a cell and apply suction to form a high resistance seal. This process isolates a small “patch” of membrane that contains one or more ion channels.
