Rat Mesentery Tissue

Rat mesentery tissue is a thin, double-layered fold of peritoneum that suspends the intestines while carrying blood vessels, lymphatic vessels, nerves, and connective tissue. Its relatively flat, transparent structure allows researchers to visualize how vascular and lymphatic networks form, remodel, and interact with surrounding extracellular matrix during development. In developmental biology, mesentery preparations support microscopy-based analysis of tissue patterning, vessel growth, inflammation, and cell migration in an intact anatomical context. These studies help link cellular behaviors to organ development and provide a model for investigating mechanisms relevant to vascular disease, regeneration, and tissue engineering.

The modified Landis technique enables paired measurement of the hydraulic conductivity of individual microvessels in the mesentery of normal and genetically modified rats under control and test conditions using microperfusion techniques. It provides a convenient method to evaluate mechanisms that regulate microvessel permeability and transvascular exchange under physiological conditions.

This article describes a simple model for stimulating angiogenesis in the rat mesentery. The model produces dramatic increases in capillary sprouting, vascular area and vascular density over a relatively short time course in a tissue that allows en face visualization of entire microvascular networks down to the single cell level.
