Hspc Differentiation Proliferation

HSPC differentiation and proliferation describe how hematopoietic stem and progenitor cells balance self-renewal with the production of increasingly specialized blood cell types, a central process in blood development and tissue maintenance. HSPCs proliferate through cell-cycle progression while signals from growth factors, cytokines, and the surrounding stem-cell niche influence whether daughter cells retain stemness or commit to myeloid or lymphoid lineages. Studying this balance helps explain embryonic and postnatal hematopoiesis, immune-system formation, and responses to injury. It also supports research on stem-cell transplantation, blood disorders, and strategies for generating defined blood cell populations in vitro.

Studying medullary thymic epithelial cells in vitro has been largely unsuccessful, as current 2D culture systems do not mimic the in vivo scenario. The 3D culture system described herein - a modified skin organotypic culture model - has proven superior in recapitulating mTEC proliferation, differentiation and maintenance of promiscuous gene expression.

In this article, we describe isolation of rhesus macaque derived adipose-derived stem cells (ADSCs) using an enzymatic tissue digestion protocol. Next, we describe ADSC proliferation which includes cell detachment, counting and plating. Lastly, ADSC differentiation is described using specific adipogenic inducing agents. Additionally, we describe staining techniques to confirm differentiation.
