Floral-dip Method

The floral-dip method is a plant transformation technique that introduces foreign DNA into developing reproductive tissues, enabling genetic modification without tissue culture. In a typical protocol, flowering plants are immersed in an Agrobacterium tumefaciens suspension containing a T-DNA vector and surfactant; the bacteria transfer T-DNA into susceptible floral tissues, and transformed seeds develop on treated plants. Researchers germinate the seeds and identify transgenic progeny using selectable markers or reporter genes, then confirm insertion and expression through molecular analysis. Widely used in Arabidopsis and other plants, the method supports gene function studies, mutant generation, promoter analysis, and crop improvement while reducing the time and specialized equipment required for conventional transformation.

This article illustrates the floral-dip method of Agrobacterium tumefaciens -mediated transformation of Arabidopsis thaliana. By introducing a cell-cycle regulated promoter-reporter, pTSO2::β-glucuronidase (GUS), into Arabidopsis, we illustrates how one detects GUS reporter expression in transgenic seedlings.

Here, we present the aluminum chloride colorimetric method, a direct analytical technique to determine flavonoids in calendula quantitatively. This approach utilizes a straightforward chemical reaction involving treating the calendula extract with an aluminum chloride reagent, forming a colored complex. The color intensity, assessed using spectrophotometry, correlates with the flavonoid concentration.

Plant Growth and Agrobacterium-mediated Floral-dip Transformation of the Extremophyte Schrenkiella parvula

Agrobacterium-mediated transformation using a floral-dip method can be successfully employed to create stable transgenic lines of the extremophyte model Schrenkiella parvula. We present a protocol modified from that for Arabidopsis thaliana, considering different growth habits and physiological characteristics of the extremophyte.

Here, we present a protocol to transform flax using Agrobacterium-mediated plant transformation via floral-dip. This protocol is simple to perform and inexpensive, yet yields a higher transformation rate than the current available methods for flax transformation.
