Immunodetection Method

An immunodetection method uses antibodies to identify and visualize specific proteins, peptides, or other antigens in a biological sample, linking molecular identity to cellular function. The method relies on selective antibody-antigen binding, followed by detection through a fluorophore, enzyme-generated color change, or chemiluminescent signal; formats include immunofluorescence, immunohistochemistry, and Western blotting. In genetics, immunodetection helps evaluate protein products of genes, compare expression patterns, determine subcellular localization, and investigate how mutations alter protein abundance or processing. These measurements complement nucleic acid analyses by showing whether genetic information produces the expected protein and functional molecular phenotype.

Described here is a method to detect and quantify mitochondrial outer membrane proteins by immunolabeling of mitochondria isolated from rodent tissue and analysis by flow cytometry. This method can be extended to assess functional aspects of mitochondrial subpopulations.
