Allogeneic T Cell Responses

Allogeneic T cell responses are immune reactions in which T lymphocytes recognize and respond to cells from a genetically different individual of the same species, making them central to transplantation immunology. They arise when donor or recipient T cells detect mismatched major histocompatibility complex (MHC) molecules or peptides presented by antigen-presenting cells through direct or indirect allorecognition, triggering cytokine release, proliferation, and cytotoxicity. These responses can promote graft rejection or graft-versus-host disease, but they may also provide beneficial graft-versus-leukemia effects after hematopoietic transplantation. Understanding their specificity and regulation supports donor matching, immunosuppressive strategies, transplant monitoring, and development of safer cellular therapies.

A rapid, simple and cost-effective protocol for the generation of donor-derived multivirus-specific CTLs (rCTL) for infusion to allogeneic hematopoietic stem cell transplant (HSCT) recipients at risk of developing CMV, Adv or EBV infections. This manufacturing process is GMP-compliant and should ensure the broader implementation of T-cell immunotherapy beyond specialized centers.

Induction of Graft Versus Host Disease in Mouse Models via Allogeneic Bone Marrow and Splenic Cell Transplantation

This video demonstrates the induction of intestinal graft-versus-host disease, GvHD, in a lethally-irradiated recipient mouse using allogeneic hematopoietic stem cells and GvHD-inducing splenic alloreactive T lymphocytes. This protocol helps establish a severe acute intestinal GvHD model to understand the mechanism of the disease and develop innovative therapeutic options.

An experimental technique for the treatment of osteochondral defects in the rabbit's knee joint is described. The implantation of allogeneic mesenchymal stem cells into osteochondral defects provides a promising development in the field of tissue engineering. The preparation of fibrin-cell-clots in vitro offers a standardized method for implantation.
