Epithelial Cell Lines

Epithelial cell lines are laboratory-maintained populations of cells derived from epithelial tissues, which form protective surfaces and regulate exchange between the body and its environment. Grown under controlled culture conditions, these cells can organize into adherent or polarized monolayers with cell junctions, surface receptors, and barrier properties that model epithelial biology. In immunology and infection research, epithelial cell lines provide reproducible systems for studying pathogen attachment, entry, replication, and interactions with host innate immune responses. They also support testing of antimicrobial or antiviral treatments and help reveal how epithelial barriers influence inflammation, tissue damage, and transmission.

We present a protocol that combines cell isolation and whole-cell patch-clamp recording to measure the electrical properties of the primary dissociated epithelial cells from the rat cauda epididymides. This protocol allows for investigation of the functional properties of primary epididymal epithelial cells to further elucidate the physiological role of the epididymis.
