Macrophage Adherence Analysis

Macrophage adherence analysis is a cell-based method that measures the ability of macrophages to attach to a culture surface, providing a practical indicator of cell recovery, activation, or phenotype in immunology and infection research. In a typical assay, macrophages are incubated on a treated surface under controlled conditions, allowing adherent cells to remain while nonadherent cells are removed by washing; the retained population is then quantified. Researchers use adherence measurements to assess macrophage behavior after exposure to pathogens, inflammatory stimuli, or experimental treatments. The results can support studies of innate immune responses, host-pathogen interactions, and changes in cellular function.

The article describes a readily easy adaptive in vitro model to investigate macrophage polarization. In the presence of GM-CSF/M-CSF, hematopoietic stem/progenitor cells from the bone marrow are directed into monocytic differentiation, followed by M1 or M2 stimulation. The activation status can be tracked by changes in cell surface antigens, gene expression and cell signaling pathways.
