Negative Bead Isolation

Negative bead isolation is a cell-separation technique that enriches a desired population by removing unwanted cells, rather than directly labeling the cells of interest. Antibody-coated magnetic beads bind specific surface markers on contaminating cells, and a magnetic field pulls the bead-bound cells away while the untouched target cells remain in suspension. In immunology and infection research, this approach can isolate immune-cell populations for flow cytometry, culture, functional assays, and pathogen-response studies. Because the target cells avoid direct bead labeling, negative selection can help preserve their surface characteristics and biological activity for downstream analysis.

DNabT cells are rare among peripheral T cells; however, they are abundant in certain non-lymphoid tissues. Difficulty of isolating DN T cells from non-lymphoid tissue hinders their functional analysis despite increasing recognized pathophysiologic significance. We describe a novel protocol for isolation of highly purified DN T cells from murine kidney.
