Host Graft Interactions

Host-graft interactions describe the biological responses that arise between a transplant recipient and the implanted tissue, organ, or cells, determining whether the graft is accepted, injured, or rejected. The process begins when the host immune system recognizes donor major histocompatibility complex molecules and other antigens, activating T cells, antibodies, inflammatory pathways, or, in some transplants, donor immune cells that attack host tissues. Understanding these interactions supports donor matching, immunosuppressive therapy, monitoring for rejection, and strategies that promote immune tolerance. This knowledge is central to improving transplant survival and developing safer approaches to tissue engineering and regenerative medicine.

High throughput validation of multiple candidate biomarkers can be performed by sequential ELISA in order to minimize freeze/thaw cycles and use of precious plasma samples. Here, we demonstrate how to sequentially perform ELISAs for six different validated plasma biomarkers1-3 of graft-versus-host disease (GVHD)4 on the same plasma sample.

Biomimetic Materials to Characterize Bacteria-host Interactions

Bacterial attachment to host cells is a key step during host colonization and infection. This protocol describes the generation of polymer-coupled recombinant adhesins as biomimetic materials which allow analysis of the contribution of individual adhesins to these processes, independent of other bacterial factors.
