Oxygen Desaturation Analysis

Oxygen desaturation analysis is the assessment of decreases in blood oxygen saturation, a clinically important measure of impaired oxygenation and respiratory function. It commonly uses pulse oximetry to track peripheral oxygen saturation over time, identifying the magnitude, duration, and frequency of drops through changes in light absorption by oxygenated and deoxygenated hemoglobin. Clinicians apply this analysis during sleep studies, exercise testing, and continuous monitoring to evaluate conditions such as sleep apnea, lung disease, and cardiovascular disorders. The results can support diagnosis, assess disease severity, guide treatment, and reveal changes in a patient’s response to oxygen therapy or other interventions.

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.

An optical system was developed to visualize hepatic microcirculation with FITC-labeled erythrocytes and to measure the partial pressure of oxygen in the microvessels with laser-assisted phosphorimetry. This method can be used to investigate physiological and pathological mechanisms by analyzing microvascular structure, diameter, blood flow velocity, and oxygen tension.
