Rat Bone Isolation

Rat bone isolation is a laboratory technique for recovering intact bone tissue from rats for controlled study of skeletal structure, cellular behavior, and disease-related changes. The procedure typically combines careful dissection with removal of surrounding muscle and connective tissue, while aseptic handling helps preserve the specimen and limit contamination; researchers may then process the isolated bone for imaging, histology, mechanical testing, or cell and marrow analysis. Murine Hind Limb Long Bone Dissection and Bone Marrow Isolation

Here we present a protocol for the dissection of hind limb long bones (femurs and tibiae) from the laboratory mouse. We further describe a rapid technique for bone marrow isolation from these bones that utilizes centrifugation for removal of bone marrow from the bone marrow space.
