Transgenic Rabbit Models

Transgenic rabbit models are rabbits genetically engineered to carry an introduced, altered, or regulated gene, providing living systems for studying gene function and human disease. Researchers typically deliver a DNA construct to embryos or reproductive cells so the sequence becomes integrated into the genome and can be inherited or expressed in selected tissues; resulting animals are then characterized by genotyping and analysis of phenotype. In medicine, these models help connect molecular changes with cardiovascular, metabolic, immune, ocular, and neurological disorders, while supporting evaluation of disease mechanisms, biomarkers, and therapies. Their size and physiology can also enable procedures and longitudinal measurements that complement mouse studies.

Recent development in gene targeting tools makes production of knockout (KO) rabbits possible. In the present work, we generated five Apolipoprotein (Apo) C-III KO rabbits using Zinc Finger Nucleases (ZFN). This work demonstrated that ZFN is a highly efficient method to produce KO rabbits.

The intensely studied nematode worm Caenorhabditis elegans can be transgenically engineered to express the human β-amyloid peptide (Aβ). Induced expression of Aβ in C. elegans muscle leads to a rapid, reproducible paralysis phenotype that can be used to monitor treatments that modulate Aβ toxicity.

This method describes the placement of interposition vein grafts in rabbits, the transduction of the grafts, and the achievement of durable transgene expression. This allows the investigation of physiological and pathological roles of transgenes and their protein products in grafted veins, and testing of gene therapies for vein graft disease.
