Transient Mcao

Transient MCAO (middle cerebral artery occlusion) is an experimental model of temporary ischemic stroke used to study how reduced cerebral blood flow and subsequent reperfusion damage brain tissue. In this method, an intraluminal filament briefly blocks the middle cerebral artery, then is withdrawn to restore blood flow and reproduce ischemia-reperfusion injury. Researchers use transient MCAO in medicine to assess infarct development, neurological deficits, neuroinflammation, vascular responses, and potential neuroprotective treatments. Because the model captures key features of reversible arterial blockage followed by reperfusion, it supports investigation of stroke mechanisms and preclinical evaluation of emerging therapies.

The intraluminal middle cerebral occlusion model in mice is herein presented. The extent of cerebral infarct is evaluated by a neurologic score and cresyl violet staining, an alternative staining to TTC, offering the great advantage to test in parallel many interest markers.

To simulate thromboembolic stroke, emboli prepared from heterologous rat blood were injected into the middle cerebral artery, followed by administration of systemic thrombolysis for recanalization. This version of the model is optimized for use in a multi-laboratory network and supports testing multiple candidate therapeutics.
