Imaging Window Implantation

Imaging window implantation is a microsurgical technique that creates stable optical access to the brain, enabling repeated visualization of neural tissue in living animals. During the procedure, a section of the skull is removed and replaced with a transparent glass or polymer window that is secured to the surrounding bone, allowing light to enter and emitted signals to be collected with microscopy. In neuroscience, this preparation supports two-photon and other fluorescence-based imaging of neuronal activity, blood flow, and cellular dynamics across days or weeks. Longitudinal observations can reveal how neural circuits change during behavior, disease progression, or treatment while reducing the need for repeated invasive surgery.

Source: Pietruczyk, E. A., et al. Laminectomy and Spinal Cord Window Implantation in the Mouse. J. Vis. Exp. (2019)This video demonstrates the implantation of a spinal cord window in an anesthetized mouse for in vivo imaging. The protocol stabilizes the exposed spinal cord using a borosilicate cover glass and a 3D-printed backplate. This method enables high-resolution two-photon microscopy to study blood-brain barrier dynamics and neurovascular interactions.

This protocol describes the fabrication of an implantable integrated imaging window using 3D laser printing. The window consists of a system of microlenses coupled with micro-scaffolds. The method involves two-photon polymerization (2PP) of the biocompatible photoresist SZ2080 in a continuous sequence, optimizing manufacturing efficiency and alignment between the different components.
