In Utero Transduction

In utero transduction is the delivery of genetic material into fetal cells before birth, enabling gene expression during nervous system development. Typically, an engineered viral vector is administered to the fetus or surrounding tissues, enters target cells, and uses cellular machinery to produce a therapeutic or experimental protein; timing, delivery route, and vector tropism influence which developing tissues are transduced. In neuroscience, this approach can model or potentially treat congenital neurological disorders by modifying neural progenitors before disease features emerge. It also supports studies of brain development, circuit formation, and early gene function while reducing barriers associated with later intervention.

Source: Lutz, D., et al. Assessment of Ultrastructural Neuroplasticity Parameters After In Utero Transduction of the Developing Mouse Brain and Spinal Cord. J. Vis. Exp. (2019).This video demonstrates viral transduction via in-utero injection in mouse embryos. A pregnant mouse is prepared for the surgery, and a midline incision is made to expose the uterine horns. A mixture of a virus containing the desired gene and a tracking dye is injected into the embryos within the uterine sacs at the...

In utero electroporation is an important technique for studying the molecular mechanisms that guide the proliferation, differentiation, migration, and maturation of cells during neural development. Electroporation enables the rapid and targeted delivery of material into cells by utilizing electrical pulses to create transient pores in cell membranes. Although electroporation has traditionally been used in in vitro studies, scientific advancements have now broadened its utilization to intact...
