Retina Isolation

Retina isolation is the laboratory process of separating the retina from surrounding ocular tissues to study its cells, circuits, and responses under controlled conditions. The procedure typically involves dissecting the eye, removing the cornea, lens, and vitreous, and carefully separating the neural retina from the retinal pigment epithelium while preserving tissue structure and viability. Isolated retinas can support electrophysiological recordings, imaging, immunohistochemistry, and molecular analyses of photoreceptors, bipolar cells, ganglion cells, and synaptic networks. In neuroscience, this preparation helps researchers investigate visual signal processing, retinal development, neurodegeneration, and responses to therapeutic interventions.

2024

This work presents an alternative flatmount retina preparation in which the removal of photoreceptor cell bodies enables faster antibody diffusion and improved patch pipette access to inner retinal neurons for immunohistochemistry, in situ hybridization, and electrophysiology experiments.

A Technique for the Prolonged Incubation of Neuronal Tissue of the Retina

2025

This video demonstrates a method for prolonged incubation of neuronal tissue of the retina. Eyecups containing retina are obtained from a mouse eye; the inner limiting membrane (ILM) is enzymatically digested to expose the neuronal cells of the retina, and either the treated eyecup or the isolated retina is transferred to a specialized incubator for prolonged incubation at a low temperature.
