Whole Mount Immunofluorescence

Whole mount immunofluorescence is a staining technique that visualizes specific proteins throughout an intact tissue or organism, preserving spatial relationships that sectioning can disrupt. The sample is fixed and permeabilized so primary antibodies can penetrate the tissue and bind target antigens, followed by fluorescently labeled secondary antibodies that enable detection with fluorescence or confocal microscopy. In neuroscience, this approach reveals neuronal architecture, glial organization, axonal projections, and protein distribution across three-dimensional specimens. By maintaining tissue continuity, whole mount immunofluorescence supports analysis of neural development, connectivity, cellular responses, and disease-related structural changes.

Quantitative Whole-mount Immunofluorescence Analysis of Cardiac Progenitor Populations in Mouse Embryos

Here, we describe a protocol for whole mount immunofluorescence and image-based quantitative volumetric analysis of early stage mouse embryos. We present this technique as a powerful approach to qualitatively and quantitatively assess cardiac structures during development, and propose that it may be widely adaptable to other organ systems.

We present a method to isolate the adult organ of Corti as three intact cochlear turns (apex, middle, and base). We also demonstrate a procedure for immunostaining with fluorescently tagged antibodies. Together these techniques allow the study of hair cells, supporting cells, and other cell types found in the cochlea.

Here, we present a protocol to quantify follicles in cultured ovaries of young mice without serial sectioning. Using whole organ immunofluorescence and tissue clearing, physical sectioning is replaced with optical sectioning. This method of sample preparation and visualization maintains organ integrity and facilitates automated quantification of specific cells.
