Capsid Variant Stratification

Capsid variant stratification is the systematic classification of viral capsid variants according to sequence, structure, and functional properties, helping researchers compare and select engineered delivery vehicles. The process combines methods such as genetic sequencing, structural analysis, and performance assays to group variants by characteristics including receptor binding, tissue tropism, particle stability, and cellular transduction. In bioengineering, this framework supports the optimization of viral vectors for gene delivery by linking capsid design features to biological outcomes. It can improve candidate selection, clarify structure–function relationships, and guide the development of vectors with more precise targeting, enhanced delivery efficiency, or reduced unwanted interactions.

Disassembly of influenza A virus cores during virus entry into host cells is a multistep process. We describe an in vitro method to analyze the early stages of viral uncoating. In this approach, velocity gradient centrifugation is used to biochemically dissect the steps that initiate uncoating under defined conditions.
