Collagen Matrigel Gel

Collagen Matrigel Gel is a three-dimensional extracellular matrix hydrogel that combines collagen, a structural protein, with Matrigel, a basement membrane protein mixture, to provide cells with a tissue-like environment for bioengineering studies. Gel formation occurs when collagen molecules assemble into fibrils under neutralizing and warming conditions, while Matrigel components polymerize into a protein-rich network, producing a hydrated scaffold with cell-adhesive cues. This composite matrix supports cell encapsulation, migration, differentiation, organoid formation, and tissue-model development. Researchers use it to investigate cell–matrix interactions, test biomaterials and therapeutics, and create in vitro systems that more closely reproduce the architecture and signaling of native tissues.

Collagen is a core component of the ECM, and provides essential cues for several cellular processes ranging from migration to differentiation and proliferation. Provided here is a protocol for embedding cells within 3D collagen hydrogels, and a more advanced technique for generating randomized or aligned collagen matrices using PDMS microchannels.

In this work, we present a technique for the rapid fabrication of living vascular tissues by direct culturing of collagen, smooth muscle cells and endothelial cells. In addition, a new protocol for the mechanical characterization of engineered vascular tissues is described.
