Microsphere Calibration

Microsphere calibration is the process of determining and standardizing the physical or measured properties of microspheres so they can serve as reliable tools in bioengineering experiments. It typically compares microspheres with known reference standards, using measurements such as size, fluorescence, density, or instrument response to identify systematic variation and establish correction factors. Calibrated microspheres improve the accuracy and reproducibility of particle characterization, imaging, flow cytometry, and assay development. They also support quality control for microsphere-based drug delivery, biomaterial testing, and diagnostic platforms, enabling researchers to distinguish true biological effects from measurement error.

Source: Laboratory of Dr. B. Jill Venton - University of Virginia Calibration curves are used to understand the instrumental response to an analyte and predict the concentration in an unknown sample. Generally, a set of standard samples are made at various concentrations with a range than includes the unknown of interest and the instrumental response at each concentration is recorded. For more accuracy and to understand the error, the response at each concentration can be repeated so an error...

We present a method for inducing elevated intraocular pressure (IOP), by injecting magnetic microspheres into the rat eye, to model glaucoma. This leads to strong pressure rises, and extensive neuronal death. This protocol is easy to perform, does not require repeat injections, and produces stable long-lasting IOP rises.
