Halocarbon Oil Overlay

Halocarbon oil overlay is a cell and embryo culture technique that places a layer of inert, hydrophobic oil over aqueous media to limit evaporation and maintain stable experimental conditions. Because the oil is immiscible with water, it forms a physical barrier that reduces air exposure and helps preserve media volume, solute concentrations, temperature, and pH during incubation, while still permitting controlled gas exchange. Researchers use halocarbon oil overlays in oocyte maturation, fertilization, embryo culture, and other small-volume biological assays. This approach improves reproducibility by protecting samples from environmental fluctuations and minimizing contamination risks.

Viral Concentration Determination Through Plaque Assays: Using Traditional and Novel Overlay Systems

Plaque assays are the gold standard for viral quantification, utilizing entrapping overlays on host cellular monolayers to determine viral titers. While various semisolid overlays have traditionally been used, here we demonstrate plaque techniques comparing semisolid overlays to a novel liquid microcrystalline cellulose among several families of viruses.
