Pathogen Exposure Assay

A pathogen exposure assay is a laboratory test that determines whether a biological sample contains evidence of prior or ongoing contact with a pathogen, supporting studies of infection, immunity, and transmission. In many formats, the assay measures pathogen-specific antibodies produced by the host, although some methods detect pathogen antigens or nucleic acids; target binding generates a measurable signal through techniques such as enzyme-linked immunosorbent assays or molecular amplification. These assays help quantify exposure in individuals or populations, evaluate immune responses, monitor outbreaks, and assess experimental infection models. Results can guide epidemiological analysis and improve understanding of host-pathogen interactions.

Single Cell Measurements of Vacuolar Rupture Caused by Intracellular Pathogens

We describe a method for tracking the endomembrane rupture elicited by the intracellular bacteria Shigella flexneri and Mycobacterium tuberculosis upon host cell invasion. Our assay makes use of CCF4, a host cytoplasmic FRET probe in live or fixed cells. This reporter is degraded by an enzyme activity present on the bacterial surface.
