Seahorse Xf 96 Analyzer

The Seahorse Xf 96 Analyzer is a 96-well extracellular flux instrument that measures cellular energy metabolism in real time, enabling comparisons of how cells produce and use energy. Its sensor cartridge detects changes in oxygen concentration and extracellular acidification above each well, while automated reagent injections perturb metabolic pathways and reveal mitochondrial respiration and glycolytic activity under defined conditions. In biology, researchers use the platform to assess basal and maximal respiration, glycolytic responses, metabolic flexibility, and drug effects in cultured cells, immune cells, and other biological models. These measurements connect cellular physiology with disease mechanisms, toxicity testing, and therapeutic development.

A description of a method for profiling mitochondrial function in cells is provided. The mitochondrial profile generated provides four parameters of mitochondrial function that can be measured in one experiment: basal respiration rate, ATP-linked respiration, proton leak, and reserve capacity.

Here, we present a protocol to perform a semi-high throughput cell migration assay on a 96-well cell culture plate. This protocol is a fast, simple and economical method to create consistent scratch wounds on a cell monolayer.

Metabolic Profile Analysis of Zebrafish Embryos

Zebrafish represent a powerful vertebrate model that has been under-utilised for metabolic studies. Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.

Blood leukocytes and platelets can be used as a marker of overall bioenergetic health of an individual and so have the potential to monitor pathological processes and the impact of treatments. Here we describe a method to isolate and measure mitochondrial function and the oxidative burst in these cells.
