Zebrafish Diabetes Model

A zebrafish diabetes model is a living experimental system that reproduces key features of diabetes, including disrupted glucose regulation and impaired insulin-producing beta-cell function, in a transparent vertebrate. Researchers generate these models through genetic manipulation or exposure to diabetogenic compounds, then monitor blood glucose, insulin signaling, pancreatic development, and beta-cell regeneration in intact animals. Because zebrafish embryos and larvae develop rapidly, remain optically accessible, and can be studied in multiwell plates, they support efficient analysis of disease mechanisms and candidate therapies. These models connect cellular and molecular findings with whole-organism physiology, while offering a practical platform for studying diabetes progression, drug responses, and pancreatic repair.

Metabolic memory is the phenomenon by which diabetic complications persist and progress unimpeded even after euglycemia is achieved pharmaceutically. Here we describe a diabetes mellitus zebrafish model which is unique in that it allows for the examination of the mitotically transmissible epigenetic components of metabolic memory in vivo.

Our protocol was developed to cleanly and easily deliver islets or cells under the kidney capsule of mice. Cells are concentrated into pellets in the final tubing used for transplanting the cells under the kidney capsule. The ease of this technique reduces stress to the cells and the mouse.

This work describes methods to establish acute and chronic hyperglycemia models in zebrafish. The aim is to investigate the impact of hyperglycemia on physiological processes, such as constitutive and injury-induced neurogenesis. The work also highlights the use of zebrafish to follow radiolabeled molecules (here, [18F]-FDG) using PET/CT.
