Partition Collection

Partition collection is the recovery of discrete reaction or sample compartments created during laboratory partitioning, preserving separated material for downstream analysis. A sample is divided into droplets, wells, or other physical compartments, which are then transferred or pooled under controlled conditions while maintaining their contents and, when required, their individual identities. In cancer research, this workflow supports compartment-specific analysis of tumor cells, nucleic acids, and treatment responses, helping researchers examine cellular heterogeneity and detect rare molecular signals. Reliable collection also improves the consistency of sequencing, imaging, and other assays that depend on well-defined sample fractions.

A technique to probe the lipid raft partitioning of fluorescent proteins at the plasma membrane of living cells is described. It takes advantage of the disparity in diffusion times of proteins located inside or outside of lipid rafts. Acquisition can be performed dynamically in control conditions or after drug addition.

Here, we present a protocol to perform a quantitative analysis of the level of plasma-membrane association for fluorescently-tagged peripherally-associated protein. The method is based on the computational decomposition of membrane and cytoplasmic component of signal observed in cells labeled with plasma membrane fluorescent marker.

We present a procedure to determine the metal-silicate partitioning of siderophile elements, emphasizing techniques that suppress the formation of metal inclusions in experiments for the noble metals. The results of these experiments are used to demonstrate the effect of core-formation on the highly siderophile element composition of the mantle.
