Fluorescein Analysis

Fluorescein analysis is the chemical measurement and characterization of fluorescein, a highly sensitive fluorescent dye used to detect and quantify substances in solution. Its xanthene-based structure absorbs light at selected wavelengths and emits longer-wavelength light, while protonation changes caused by pH strongly influence its color, fluorescence intensity, and absorption spectrum. Chemists use spectrophotometry and fluorescence spectroscopy to determine fluorescein concentration, evaluate acid-base behavior, and monitor interactions with solvents or other chemical species. These measurements support quantitative analysis, tracer studies, environmental monitoring, and the development of fluorescent probes for analytical and biochemical research.

In the present study, fluorescein-isothiocyanate-labeled (FITC) dextran is administered to mice via oral gavage to evaluate intestinal permeability both in vivo and in plasma and fecal samples. As gut barrier function is affected in many disease processes, this direct and quantitative assay can be used in diverse research areas.
