Valve Closure

Valve closure is the sealing of a valve opening to control fluid movement, a fundamental process in cardiovascular function and clinical assessment. In the heart, valves close when pressure changes across their leaflets or cusps reverse the direction of the pressure gradient: atrioventricular valves shut during ventricular contraction, while semilunar valves close as the ventricles relax. Proper closure prevents regurgitation and supports efficient, one-way blood flow. Abnormal closure can produce murmurs, volume overload, or reduced cardiac output, making valve motion an important focus of physical examination, echocardiography, and the evaluation of conditions such as stenosis and insufficiency.

This protocol describes the isolation, culture, and calcification of rat-derived valve interstitial cells, a highly physiological in vitro model of calcific aortic valve disease (CAVD). Exploitation of this rat model facilitates CAVD research in exploring the cell and molecular mechanisms that underlie this complex pathological process.
