Cd31 Cd45

CD31 and CD45 are cell-surface markers used to identify and distinguish endothelial and hematopoietic populations, making them valuable tools in developmental biology. CD31, also called PECAM-1, is enriched on endothelial cells and some blood cells, whereas CD45 is a pan-leukocyte antigen expressed by hematopoietic cells; fluorescent antibodies against these proteins enable characterization by flow cytometry or microscopy. Their combined expression helps researchers separate endothelial, hematopoietic, and transitional populations during embryonic blood development, including studies of the endothelial-to-hematopoietic transition. CD31/CD45 profiling supports cell isolation, lineage analysis, and investigation of how vascular and blood systems develop.

The differentiation of white and beige adipocytes from adipose tissue vascular progenitors bears potential for metabolic improvement in obesity. We describe protocols for a CD34+CD31+ endothelial cell isolation from human fat and for a subsequent in vitro expansion and differentiation into white and beige adipocytes. Several downstream applications are discussed.

Liver injuries are accompanied by progenitor cell expansion that represents a heterogeneous cell population. Novel classification of this cellular compartment allows for the distinguishing of multiple subsets. The method described here illustrates the flow cytometry analysis and high purity isolation of various subsets that can be used for further assays.

Hematopoietic stem and progenitor cells (HSPC) derive from specialized (hemogenic) endothelial cells during development, yet little is known about the process by which some endothelial cells specify to become blood forming. We demonstrate a flow-cytometry based method allowing simultaneous isolation of hemogenic endothelial cells and HSPC from murine embryonic tissues.
