Cd11b Cells

CD11b cells are immune cells that express CD11b, the αM subunit of the β2 integrin Mac-1, and are important markers of myeloid-cell identity and activation. CD11b pairs with CD18 to form integrin receptors that support adhesion, migration, complement recognition, and phagocytosis as cells respond to tissue damage or infection. In immunology, CD11b is commonly detected by flow cytometry or immunostaining to identify and characterize monocytes, macrophages, neutrophils, and other myeloid populations. Measuring CD11b expression helps researchers track innate immune recruitment, examine host–pathogen interactions, and assess inflammatory or immunosuppressive cell responses.

Here, we present a protocol to isolate microglia from postnatal mouse pups (day 1) for in vitro experimentation. This improvised method of isolation generates both high yield and purity, a significant advantage over alternate methods that allows broad range experimentation for the purposes of elucidating microglial biology.

Inflammation plays a central role in the pathogenesis of ischemic stroke. Increasing evidence suggests that it acts as a double-edged sword which exacerbates early brain injury, but also contributes to later repair. This protocol describes the isolation of immune cells from the ischemic brain and their subsequent flow cytometric phenotyping.

In Vitro Generation of Plasmacytoid Dendritic Cells from Common Lymphoid Progenitors

This video demonstrates an in vitro assay to generate plasmacytoid dendritic cells (pDCs) from common lymphoid progenitor cells (CLPs). Upon co-culturing purified CLPs with γ-irradiated feeder cells in the presence of Fms-like tyrosine kinase-3 ligand (FL), the CLPs differentiate into non-adherent pDCs, the presence of which is confirmed using flow cytometry.

Source: Hidmark, A. S., et al. Analysis of Immune Cells in Single Sciatic Nerves and Dorsal Root Ganglion from a Single Mouse Using Flow Cytometry. J. Vis. Exp. (2017). This video demonstrates the use of flow cytometry to analyze immune cells in mouse sciatic nerves and dorsal root ganglia (DRGs). The process involves labeling immune cells with fluorophore- and biotin-tagged antibodies, stabilizing and permeabilizing the cells, and using fluorescent markers to detect cell-surface and nuclear...

Isolating Microglia from Mouse Brain Demyelinating Lesions Using Magnetic Activated Cell Sorting

This video demonstrates a protocol for extracting microglial cells from demyelinating lesions in mouse brains using magnetic cell sorting. The protocol involves dissecting the lesioned brain tissue containing microglial cells, treating the tissue with specific enzymes to obtain a single-cell suspension, and utilizing microglial receptor-specific magnetic beads to isolate the microglial cells.
