Footpad Swelling Measurement

Footpad swelling measurement is a quantitative method for assessing localized tissue inflammation, commonly in experimental immunology and infection studies. After an antigen, pathogen, or inflammatory stimulus is introduced into the footpad, immune-cell recruitment and vascular changes increase fluid accumulation, causing measurable tissue enlargement; researchers typically track this response with calipers or a volume-displacement instrument over time. Changes in thickness or volume provide an accessible indicator of the magnitude and timing of inflammation, supporting evaluations of cellular immune responses, host-pathogen interactions, vaccine efficacy, and anti-inflammatory treatments. Standardized measurements also enable comparisons among experimental groups and help relate visible tissue changes to underlying immune mechanisms.

Forskolin-induced Swelling in Intestinal Organoids: An In Vitro Assay for Assessing Drug Response in Cystic Fibrosis Patients

This protocol describes an assay for measuring CFTR function and CFTR modulator responses in cultured tissue from subjects with cystic fibrosis (CF). Biopsy-derived intestinal organoids swell in a cAMP-driven fashion, a response that is defective (or strongly reduced) in CF organoids and can be restored by exposure to CFTR modulators.

The present protocol describes a unique, clinically relevant model of peripheral arterial disease that combines femoral artery and vein electrocoagulation with the administration of a nitric oxide synthase inhibitor to induce hindlimb gangrene in FVB mice. Intracardiac DiI perfusion is then used for high-resolution, three-dimensional imaging of the footpad vasculature.

We describe a valuable diagnostic assay that could potentially be used to decide the withdrawal of immunosuppression after transplant without elevated risk of graft rejection. The assay uses the principles of Delayed Type Hypersensitivity and provides accurate assessment of both donor specific effector and regulatory immune responses mounted by recipients.
