Vascular Stem Cells

Vascular stem cells are self-renewing or progenitor-like cells associated with blood vessels that can contribute to vascular growth, maintenance, and repair, making them important in cardiovascular medicine. Through controlled proliferation and differentiation, they may generate endothelial cells that line vessels or vascular smooth muscle cells that support vessel structure, responding to local signals such as injury and tissue environment. Studying these cells helps clarify how vessels develop and remodel and may support applications in cardiovascular disease modeling, tissue engineering, and regenerative therapies. Their biology also informs strategies to restore blood flow and improve vascular graft design.

A Novel Mammary Fat Pad Transplantation Technique to Visualize the Vessel Generation of Vascular Endothelial Stem Cells

This work demonstrates a novel approach to assess the proliferation, differentiation, and vessel-forming potential of vascular endothelial stem cells (VESCs) through mammary fat pad transplantation followed by whole-mount tissue preparation for microscopic observation. A lineage tracing strategy to investigate the behavior of VESCs in vivo is also presented.

Derivation of Hematopoietic Stem Cells from Murine Embryonic Stem Cells

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

To our knowledge, systematic and efficient methods to transfect human induced pluripotent stem cells (iPSCs) have not been reported.
