Apical Dendritic Complexity

Apical dendritic complexity is the structural organization of branching, length, thickness, and synaptic features within a neuron's apical dendrite, a major determinant of how that cell receives and integrates information. In cortical pyramidal neurons, the apical arbor extends toward superficial layers, where its branches collect inputs from diverse sources; branch geometry and membrane properties shape signal propagation, local integration, and the influence of distal synapses on somatic output. Researchers quantify this complexity through morphological tracing, reconstruction, and measures such as branch number or total length to compare development, plasticity, disease-related changes, and circuit function.

This video demonstrates a protocol for assessing the activation of dendritic cells using immunoglobulin G (IgG)-coated tumor cells. Dendritic cells internalize the IgG-coated tumor cells, process the tumor antigens, and present them on the surface through MHC-II molecules, along with the co-expression of the costimulatory molecule CD86. These dendritic cells then undergo MHC-II and CD86-targeted immunostaining and flow cytometry analysis to identify their activation state.
