C-fos Staining

C-fos staining is an immunohistochemical method used in neuroscience to identify cells that have recently undergone increased activity, making it useful for mapping functional responses in nervous tissue. Neural stimulation can induce transcription of the immediate-early gene c-fos, followed by production of c-Fos protein; antibodies against this protein then reveal labeled nuclei in fixed brain sections. Researchers use c-fos staining to associate specific brain regions and neuronal populations with sensory processing, behavior, stress, learning, and disease-related activity. Because c-Fos expression develops over minutes to hours and reflects cellular activation indirectly, the method provides spatial information but limited temporal resolution.

The goal of this study is to identify reward-related distributed brain networks by delineating a reliable immunohistological technique using cellular c-fos activation to measure simultaneous changes in dopamine pathways and terminal sites after novel ingestion of fat and sugar in rats.

Here we present a Fluorescence Activated Cell Sorting (FACS) protocol to study molecular alterations in Fos-expressing neuronal ensembles from both fresh and frozen brain tissue. The use of frozen tissue allows FACS isolation of many brain areas over multiple sessions to maximize the use of valuable animal subjects.

Here, we describe protocols for the genetic and chemical validation of c-Fos and Dusp1 as a drug target in leukemia using in vitro and in vivo genetic and humanized mouse models. This method can be applied to any target for genetic validation and therapeutic development.
