Cell Attached Recording

Cell-attached recording is an electrophysiological technique that measures electrical activity from an intact cell without rupturing its membrane, making it valuable for studying neuronal signaling. In this configuration, a fine glass micropipette forms a high-resistance seal with the cell membrane, allowing the recording of ion-channel currents or action-potential activity from the membrane patch beneath the pipette. Because the cell remains largely undisturbed, researchers can examine spontaneous firing, synaptic responses, and channel behavior under near-physiological conditions. In neuroscience, cell-attached recordings help characterize neuronal excitability, identify functional cell types, and investigate how drugs, circuits, or disease-related changes alter electrical communication.

Described here is a procedure for obtaining long stretches of current recording from one ion channel with the cell-attached patch-clamp technique. This method allows for observing, in real time, the pattern of open-close channel conformations that underlie the biological signal. These data inform about channel properties in undisturbed biological membranes.

Here we describe a quick and simple method to measure cell stiffness. The general principle of this approach is to measure membrane deformation in response to well-defined negative pressure applied through a micropipette to the cell surface.
