Cortical Slice Preparation

Cortical slice preparation is a neuroscience technique that produces thin, viable sections of cerebral cortex for controlled study outside the intact brain. Typically, freshly isolated tissue is stabilized in chilled, oxygenated artificial cerebrospinal fluid and sectioned with a vibratome or similar instrument to preserve cellular structure and local synaptic connections. Researchers use these slices for electrophysiology, calcium imaging, pharmacology, and microscopy, allowing precise analysis of neuronal activity, circuit connectivity, and responses to drugs or stimulation. Because the preparation provides experimental access while retaining elements of native cortical organization, it supports studies of development, sensory processing, plasticity, and neurological disease mechanisms.

Preparation of Dissociated Mouse Cortical Neuron Cultures

This video shows a procedure for generating neuronal cultures from late embryo and early postnatal mouse cortex. The present method allows reproducible cryostat sectioning of small, difficult-to-manage, tissue pieces, such as biopsies and brain slices. We utilize a simple aluminum freezing stage to facilitate handling of tissue and a standard cryostat to routinely produce 5-10 micron serial sections from 400 micron thick brain slices.
